The Certificate of Analysis (CoA) is one of the most important documents in pharmaceutical trade. Every shipment of medicines should be accompanied by a CoA for the specific batch being shipped — and understanding what it contains, and how to assess it, is a core competency for pharmaceutical import buyers.

What Is a CoA?

A CoA is a document issued by the manufacturer's Quality Control (QC) department, certifying that a specific batch of product has been tested and found to conform to its specifications. It is a batch-specific document — each batch number has its own CoA.

The CoA bridges the gap between what you ordered ("Amoxicillin 500mg capsules meeting IP specifications") and what you received ("Batch No. XYZ of Amoxicillin 500mg capsules, tested and confirmed to meet IP specifications").

Standard CoA Elements

A well-prepared pharmaceutical CoA from an Indian manufacturer will contain:

1. Product Identification

- Product name: Brand name (if branded) and/or INN + strength + dosage form - Manufacturer name and address - Manufacturing licence number

2. Batch Information

- Batch number / Lot number: The unique identifier for this production lot — use this to match the CoA to your physical shipment - Batch size: How many units were produced in this batch - Date of manufacture: Month and year - Date of expiry (Expiry date): The end of the stated shelf life

Verify that the batch number on the CoA matches the batch number on the actual product packaging. A mismatch means the CoA is not valid for the goods you received.

3. Test Parameters, Specifications, and Results

This is the substantive section. For each test performed, the CoA states:

Key tests to look at: Description: Visual/physical description of the product (colour, shape, coating). Should match what you receive. Identification: Confirms the product contains the declared active ingredient (typically by IR spectroscopy, HPLC, or chemical test). Must show "Complies" or "Positive." Assay (Potency): The percentage of active ingredient found versus the label claim. For a 500mg capsule, an assay of 99.5% means the actual content is approximately 497.5mg per capsule. Specifications are typically ±5% of label claim (95–105%), or tighter for narrow-therapeutic-index drugs. Assay results near the limits of specification are technically passing but worth noting. Dissolution (for oral solid dosage forms): Measures how much of the active ingredient dissolves in a specified time under defined conditions — a surrogate for bioavailability. Specification is typically expressed as "Not less than X% dissolved in Y minutes." Moisture content / Loss on drying: Particularly important for hygroscopic products and capsule fills. High moisture can indicate improper storage or manufacturing. Microbial limits: For non-sterile products, confirms that microbial contamination is within acceptable limits. For sterile products (injectables), the test is "Sterility" — a pass/fail test confirming absence of viable microorganisms. Related substances / Impurities: Tests for degradation products and process-related impurities. Each impurity should be below specified limits.

4. Status and Approval

- Status: Should state "Complies" or "Passes" for each test - Authorised signatory: The QC Manager or Quality Assurance head who signs and dates the release. The CoA should bear an authorised signature and the manufacturer's official stamp/letterhead.

Red Flags to Watch For

- Batch number mismatch between CoA and physical product - Results at or very close to specification limits (technically passing, but indicating quality edge cases) - Missing tests that you would expect for the product type (e.g., dissolution not reported for an oral solid) - CoA on trader's letterhead rather than manufacturer's — the CoA must come from the manufacturer's QC department; a trader re-issuing a CoA on their own letterhead is not acceptable - Vague or generic results ("within limits" without actual measured values) — a proper CoA shows actual numeric results - Expired or pre-signed CoA (date of issue should match or follow the batch release date)

Using the CoA in Your Import Process

1. Request the CoA from your supplier before or at the time of shipment 2. Match the batch number on the CoA to the batch number on the product packaging/outer carton 3. Review key parameters — at minimum: identification, assay, expiry date 4. File the CoA with your import records — it is the primary quality document for the batch
